This document introduces high capacity affinity depletion column technology, which can highly specifically deplete the top six high-abundance proteins in human serum, which is advantageous for the detection and identification of low-abundance proteins.
This document introduces the Agilent High Capacity Multiple Affinity Removal Column, an improved version of the Multiple Affinity Removal approach designed for proteomics applications. The column utilizes a novel attachment process for affinity binders, including polyclonal IgG-based antibodies, to specifically remove high-abundant proteins such as HSA, IgG, haptoglobin, transferrin, IgA, and alpha-1-antitrypsin from complex biological fluids like human serum, plasma, and cerebral spinal fluid.
| Targeted proteins | HSA, IgG, haptoglobin, transferrin, IgA, and alpha-1-antitrypsin |
|---|---|
| Depletion efficiency | Greater than 99% |
| Column capacity (4.6 x 50 mm) | At least 20 uL of undiluted serum |
| Column capacity (4.6 x 100 mm) | At least 40 uL of undiluted serum |
| Reproducibility | At least 300 injections |
The column depletes HSA, IgG, haptoglobin, transferrin, IgA, and alpha-1-antitrypsin.
The column achieves greater than 99% depletion of the targeted proteins.
The high-capacity depletion column provides robust performance for at least 300 injections of human serum.
