This application note describes how the Agilent 2100 bioanalyzer can be used for the detection of a point mutation in the human prothrombin gene. The prothrombin protein plays a key role in blood clotting. A single nucleotide polymorphism (SNP), i.e. a point mutation in the prothrombin gene results in a common hereditary predisposition to venous thrombosis. We present a convenient and reliable mutation detection method by a PCR-based assay including a specific restriction digest (PCR-RFLP). Due to its high sensitivity and resolution, the applied microfluidic assay proves to be extremely suitable for detec- tion of the obtained PCR products and restriction fragments. The developed method is suitable for the workflow at Laborigo molecular diagnostic laboratory.

The 5975 inert MSD – Benefits of Enhancements in Chemical Ionization Operation Technical Note This Technical Note describes the complete integration of chemical ionization (CI) on the 5975 inert MSD (mass selective detector), which now makes all the power of CI mass spectrometry (MS) conveniently available. CI has been fully automated by providing auto-tuning with electronic control of the reagent gases. All modes of MS operation in a single sequence, completely free of operator intervention, are now possible. Analytes can be sensitively determined in samples by implementing alternating positive- or negative-CI (PCI or NCI) modes. On the 5975 inert MSD, CI is as easy as EI (electron impact ionization).

A USGS analytical method using gas chromatography/mass selective detector (GC/MSD) was modified and used to identify the source of pesticides and to calculate the pesticide load in river water during storm seasons. The method was validated to cover 12 organophosphate pesticides and 5 pyrithroids. The limits of quantitation for these target compounds are 10–500 ppt using a GC/MSD selected ion monitoring (SIM) method.

The use of a highly inert capillary column can facilitate the quantitation of low levels of very active compounds. The use of the highly inert HP-5MSi column in conjunction with the Agilent 6890/5973 inert gas chromatograph/mass spectrometer (GC/MS) system allows the measurement of pesticides and environmental contaminants at low levels while maintaining response linearity over a wide concentration range.

This document introduces the optimized sources for electron ionization (EI) and chemical ionization (CI) of Agilent 5973x gas chromatograph/mass selective detectors (GC/MSDs). It also demonstrates how to acquire high-quality EI spectra with the CI source without changing the sources. The experimental results show that using the CI source can obtain comparable major and minor peaks to the EI source.
