Technical manual for Promega's MSI Analysis System (Version 1.2) describing components, storage, DNA extraction, PCR setup and data analysis.
The MSI Analysis System (Version 1.2) is a fluorescence‑based multiplex PCR kit for detecting microsatellite instability in human DNA. The manual provides an overview of the system, the seven microsatellite markers, internal lane standard ILS‑600, and the workflow from DNA extraction to result interpretation.
Detailed procedures cover reagent preparation, PCR cycling conditions, capillary electrophoresis on ABI PRISM® instruments, matrix generation, and troubleshooting. Appendices list related Promega products and allele frequency tables for the single‑base repeat loci. This manual is written in Chinese.
| Reactions per kit | 100 reactions (50 pairs) |
|---|---|
| Storage temperature | -30 °C to -10 °C |
| Light sensitivity | MSI 10 X Primer Pair Mix and ILS‑600 must be protected from light |
Each kit contains enough reagents for 100 reactions (50 paired reactions).
All components should be stored between –30 °C and –10 °C.
A thermal cycler such as GeneAmp® System 9600 or 9700 (Applied Biosystems) is required.
