This application note describes a method for analyzing oxycodone and its metabolites (noroxycodone, oxymorphone, and noroxymorphone) in plasma using high-performance liquid chromatography/mass spectrometry (HPLC/MS) and the Agilent ZORBAX Rapid Resolution High Throughput (RRHT) StableBond SB-C18 column for chromatographic separation. The method employs an ammonium acetate/acetonitrile gradient and detection by electrospray positive ionization mode mass spectrometry. Spiked human plasma samples are subjected to solid-phase extraction prior to HPLC/MS analysis. The method provides fast, efficient analysis with minimal solvent consumption, which increases productivity, and good linearity (R2 > 0.9900) and reproducibility (less than 10% difference between replicate analyses) for all compounds.
This document details a method for analyzing oxycodone and its oxidation metabolites (noroxycodone, oxymorphone, noroxymorphone) in human plasma. The method combines high-speed liquid chromatography with mass spectrometry detection using an Agilent ZORBAX Rapid Resolution High Throughput StableBond SB-C18 column.
The procedure involves solid-phase extraction of spiked human plasma samples followed by analysis via LC/MS in electrospray positive ion mode. The method achieves good linearity (R² > 0.9900) and reproducibility, with a limit of detection for oxycodone at 0.5 ng/mL. This manual is written in Japanese.
The detection and quantification limit for oxycodone is 0.5 ng/mL.
The column measures 2.1 mm in inner diameter and 50 mm in length.
The difference between sets of replicate samples must be less than 10%.
