This document describes a simple isocratic method for analyzing artemisinin using the Agilent 1200SL system with high-speed separation high-throughput (RRHT) columns and ChromSword software. The method development was automated using the ChromSword software, which helped in selecting the optimal column. By utilizing a fully automated unmanned method development program, the analysis time was significantly reduced.
This document details a study on the automated analysis of artemisinin, a key active ingredient in Artemisia annua used to treat malaria. The authors utilized an isocratic method developed with minimal manual intervention by combining high-speed separation (RRHT) columns with Agilent's ChromSword software for automation.
The experiment employed an Agilent 1200 Series HPLC system equipped with a G1312B Binary Pump, G1376C Autosampler, G1316B Temperature Control Compartment, and G1316C Diode Array Detector. The study successfully separated artemisinin, artesunate, and two DHA epimers within 6 hours using the ZORBAX Eclipse Plus C18 column (4.6 mm x 100 mm, 1.8 µm) and a mobile phase of 0.1% formic acid in water and acetonitrile. This manual is written in Japanese.
The automated experiment analysis was executed over a period of 6 hours.
The lowest value for the separation factor observed in the experiment was 256.
