This document describes a method for detecting Hypericin and provides an application example of the method.
This document presents a method developed for the separation of hypericin, a major component in the herbal extract of Hypericum Perforatum (St. John's Wort). The method was created in response to increasing interest in natural product therapies and growing government regulation concerns.
The procedure utilizes an Agilent ZORBAX XDB-C8 column with a mobile phase consisting of 23% 25 mM Na2HPO4 (pH 7.0) and 77% Methanol. Key highlights include good peak shape for hypericin at neutral pH, extended column lifetime due to double endcapping, and adequate separation from other UV-absorbing compounds.
| Column | ZORBAX XDB-C8 (4.6 x 150 mm) |
|---|---|
| Mobile Phase | 23% 25 mM Na2HPO4, Dibasic (pH 7.0) : 77% Methanol |
| Injection Volume | 2 uL |
| Temperature | 35°C |
| Detection | UV (254 nm) |
A method was developed for hypericin and an herbal product of St. John's Wort due to increasing interest in natural product therapies.
The method provides good peak shape for hypericin at neutral pH, longer column lifetime, and adequate separation from other UV-absorbing compounds.
