About this manual

The Absolutely RNA 96 Microprep Kit allows for the rapid purification of high-quality total RNA from small samples of cultured cells. The method eliminates toxic phenol-chloroform extractions and time-consuming ethanol precipitations by using a lysis buffer containing guanidine thiocyanate to lyse cells and prevent RNA degradation.

Following cell lysis, the sample is transferred to a 96-well binding plate with a silica-based fiber matrix where RNA binds. Treatment with low-salt wash buffers followed by a DNase digestion step removes contaminating DNA and proteins. Highly pure RNA is finally eluted from the fiber matrix into a collection plate, suitable for downstream applications like cDNA synthesis and PCR.
