About this manual

The Agilent miRNA 1st-Strand cDNA Synthesis Kit provides reagents to elongate miRNAs in a polyadenylation reaction using E. coli Poly A Polymerase (PAP) and then reverse transcribe the polyadenylated RNA into QPCR-ready cDNA. The kit includes specific enzymes, buffers, primers, and dNTPs necessary for these steps.

Frequently asked questions

What is the optimal RNA purity ratio?

RNA samples with OD260/280 ratios of 1.8-2.0 are optimally pure.

Is enrichment for miRNA necessary?

No, enriching the RNA sample for miRNA is not necessary but may improve detection of some difficult targets.

What should be done if Manganese Chloride is used?

If MnCl2 is included in the polyadenylation reaction, purify the polyadenylated RNA with a phenol-chloroform extraction and ethanol precipitation before cDNA synthesis.
