This article describes the quantification of potential genotoxic impurity D in Atenolol using LC/ESI/MS/MS method with Agilent 1200 Series RRLC and 6410B Triple Quadrupole LC/MS instruments. The developed method is simple, shorter in runtime, with better resolution and higher sensitivity than the existing European and United States Pharmacopeia methods. Agilent's MassHunter Optimization software was used for quick optimization of MS parameters. By connecting the Agilent 6410B Triple Quadrupole (QQQ) LC/MS detector to an Agilent 1200 Series RRLC system, the quantification range can be effectively reduced further down to 5 ppb with a linearity coefficient greater than 0.9992.

Quantitative proteomics using multiple reaction monitoring (MRM) has emerged as an important methodology for biomarker validation. Multiple-reaction monitoring on a triple quadruple (QQQ) mass spectrometer provides superior sensitivity and selec- tivity for targeted peptides in a complex sample. MRM also offers high precision in quantitation and a fast scan speed, which makes it an ideal technology for validating biomarkers in a high-throughput fashion. This application note demonstrates the entire workflow for biomarker validation offered by Agilent Technologies using multiple reaction monitoring together with statistical analysis software. Thousands of peptide transitions were generated using Agilent Q-TOF and Spectrum Mill MRM selector. The transition list was imported into MassHunter acquisition software equipped with dynamic MRM functionality and monitored in a single LC/MS run. Finally, MRM quantitation results were analyzed using Mass Profiler Pro for principle component analysis (PCA), hierarchical clustering and ANOVA analysis.

Agilent has further improved the sample throughput capabilities of its 7500cx ICP-MS with Octopole Reaction System (ORS) using a newly configured Integrated Sample Introduction System–Discrete Sampling (ISIS-DS) accessory, and helium collision mode. Employing this new methodology, a complete suite of 30 or more elements can be analyzed in compliance with USEPA criteria (spectrum mode, three replicates, and sub-ppb MDLs) in approximately 75 seconds, sample to sample, with excellent removal of polyatomic interferences. This Application Note describes a rapid method for analysis of polynuclear aromatic hydrocarbons (PAHs) in soil and sediment samples by GC-MS using an Agilent Low Thermal Mass (LTM) GC/MS system and Capillary Flow Technology QuickSwap for Backflush. The cycle time for GC-MS analysis of PAHs was dramatically improved through the combined use of a narrow-bore (180-µm id) column installed in an LTM module and capillary column back-flushing with QuickSwap. Improvements were achieved while maintaining resolution. Agilent Capillary Flow Technology enabled column backflushing to remove low-volatili- ty material from the capillary column, reducing bakeout and maintenance time. The method resulted in increased sample throughput and lab productivity, while maintain- ing analytical performance.

This application note is targeted for ultra-high productivity of total petroleum hydro- carbons (TPH) analysis in environmental laboratories. Agilent's Low Thermal Mass (LTM) technology is employed here to perform ultra-fast gas chromatographic (GC) separations. The LTM technology uses a column module combining a fused silica cap- illary column with heating and temperature-sensing components wound around it, which can be heated and cooled very efficiently. In this application note, the speed of analysis for the hydrocarbon group eluting between C10 and C44 can be dramatically increased to about 13 times faster than a conventional method. In addition, the ultra- fast cooling function of an LTM module can reduce the total GC cycle time to 5.1 minutes. The simultaneous dual-tower injection from Agilent is used to further double productivity. The final result for TPH analysis productivity is 5.1 minutes per two samples.
