The document describes the use of the Agilent 6550 iFunnel Q-TOF LC/MS system for high-throughput, high-efficiency metabolome profiling. Compared to the previous generation Q-TOF instrument, this platform has increased sensitivity and resolution, resulting in the detection of an order of magnitude more ions. It provides a 6 to 7-fold increase in coverage of the E. coli metabolome and more detailed pathway analysis.

This document describes the analysis of BTEX in natural water using SPME technique. BTEX are monocyclic aromatic hydrocarbons with moderate solubility. They are constituents of mineral oil products and widely used as solvents in industrial processes. Additionally, they are major water-soluble constituents of petroleum derivatives (gasoline). The author used GC with FID for analysis and provided detailed information on the instrumentation and conditions.

This application note provides a method for comprehensive detection of pesticides in vegetable juice matrices using GC/MS and LC/MS methods. Methods have been developed on Agilent GC/MS and LC/MS instrument platforms that reliably detect and quantitate 39 pesticides in a vegetable juice matrix. Most of the limits of quantitation (LOQ) values for GC/MS SIM ranged from 5 to 50 ng/mL, compared with 0.25 ng/mL to 10 ng/mL for GC/MS/MS, and 0.1 ng/mL to 10 ng/mL for LC/MS/MS. Both sensitivity and selectivity are improved using MS/MS and recovery of most spiked pesticides ranged from 70 to 120%.

An analytical method for screening and confirming the presence of 84 pharmaceuti- cals and 23 labeled standards in water samples for a total of 107 components was developed using the Agilent 6460 Triple Quadrupole Mass Spectrometer (Triple Quad MS) with Jet Stream Technology.

This application note demonstrates sensitive and reproducible HPLC analysis of N-linked glycans of human immunoglobulin G using the Agilent 1260 Infinity Bio-inert Quaternary LC System with fluorescence detection. The precision of retention times and areas was determined for analysis of a N-linked human IgG glycan library. In addition, good linearity in a range from 0.008 to 1 pmol was determined for a mix of five glycan standards (Man5, G2, G2S1, G2FS1, and G2S2) with low limits of detection and quantification. The Agilent 1260 Infinity Bio-inert Quaternary LC System with fluorescence detection is an optimal system for sensitive and reproducible analysis of 2-AB derivatized N-glycans released from human immunoglobulin G.

This Application Note shows that unwanted retention and peak tailing of adenosine triphosphate can be completely prevented when using the Agilent 1260 Infi nity Bio-inert Quaternary LC System. Due to the iron and steel-free design of the Agilent 1260 Infi nity Bio-inert Quaternary LC System, phosphate compounds can be analyzed without any issues regarding the formation of phosphate-iron complexes found with stainless steel systems. Even in the presence of high organic mobile phases, good peak shapes were observed without peak tailing or area reduction.

A method for simultaneous determination of malachite green and crystal violet, and their metabolites leuco-malachite green and leuco-crystal violet in fish, was developed and validated. The analytes were extracted by solid phase extraction and quantified by liquid chromatography coupled to electrospray ionization tandem mass spectrometry operating in positive ion multiple reaction monitoring mode. The method provided a 0.5 ng/g limit of quantification for all these compounds in fish. The dynamic calibration ranges for these compounds were obtained from 0.5 to 100 ng/g. The overall recoveries ranged from 96 to 109% with RSD values between 1.7 and 4.5%.

This application note describes a rapid method for detecting benzoylecgonine (BZE), the major metabolite of cocaine, in urine using the Agilent RapidFire High-throughput Mass Spectrometry System. Results were achieved with much faster sample cycle times and similar analytical output compared to GC/MS or LC/MS assays.

This application note describes a method using high resolution accurate mass (HRAM) LC/MS for the quantitation of cystine and related metabolites in white blood cells. Excellent assay performance was achieved for cystine quantitation in white blood cells (WBCs) using the ultra-high resolving power and mass accuracy of an Agilent 6530 Accurate-Mass Q-TOF LC/MS System. Further, related metabolites were successfully identified and quantitatively profiled. The HRAM LC/MS data acquired in this study can be retrospectively analyzed to search for more metabolites and biomarkers without sample re-injection.
