Mercury is one of the more toxic elements to living organisms. It is encountered at relatively low concentrations in the environment but with its amplification through the food chain, the final concentration in some foods can be relatively high. In addition, its toxicity is not only linked to its total concentration but is also dependent on the species. Mercury speciation in food analysis is necessary to fully estimate the toxic potential of this element to humans. Due to the low detection limits required for the analysis of Hg species, GC-ICP-MS has been the preferred method. However, with the sensitivity improvements in modern ICP-MS systems, the use of HPLC has become practical for the separation. Coupling HPLC and ICP-MS is particularly attractive due to the simple and fast connection between the techniques. Furthermore, in contrast to a GC analysis, sample extracts are directly injected into the system and no species derivatization is required, greatly simplifying the sample preparation and reducing the cost of the total analytical procedure.

To ensure food safety and safeguard human health, the characterization of the elemental composition of a wide range of food types is required. The possibility of measuring all the required elements with a single configuration using the Agilent 7700x ICP-MS is investigated. The determination of trace and major elements simultaneously is possible due to the 9 orders of magnitude provided by the detector. This application note describes an ultrafast SPE/MS/MS method, using the Agilent RapidFire High-Throughput Mass Spectrometry System, with a sample analysis time of <10 seconds per sample, for the analysis of levetiracetam in human serum. The Agilent RapidFire/MS System exhibited comparable accuracy, precision, linearity, and sensitivity to LC/MS/MS, but with a sample analysis cycle time 10-fold faster than LC/MS/MS.

This study focuses on the rapid screening of an entire hippocampus section of mouse brain for the study of Alzheimer’s disease. This Application Note describes the performance comparison and benchmark of the Agilent 2200 TapeStation system with the Agilent 2100 Bioanalyzer system, the industry standard for RNA quality control. The 2200 TapeStation system performs automated analysis of up to 96 samples with approximate analysis time of 1 minute per sample. This Application Note demonstrates the high correlation between RINe generated by the 2200 TapeStation system with the RIN quality assessment of the 2100 Bioanalyzer system. The high reproducibility of RINe achieved with the 2200 TapeStation system in RNA quality assessment is also shown. The 2200 TapeStation system is, therefore, a reliable system for RNA quality control.

Gene expression analysis is currently carried out using three methods that provide complementary data: microarrays, RNA-Seq, and RT-qPCR. Microarrays provide comprehensive whole-genome coverage in single experiments, with rapid data acquisition and results at a low price per sample.

This application note demonstrates the concordance in the results achieved with identical oligonucleotide probes on two versions of Agilent miRNA microarrays [(AMADID 021827, release 12.0) and (AMADID 031181, release 16.0)], based on different revisions of the miRBase database and provided on different microarray formats. Since probes for a given microarray will remain constant for subsequent versions of Agilent SurePrint G3 Human miRNA microarrays, barring any miRBase sequence changes, it is necessary to confirm that the TotalGeneSignal achieved is consistent. This application note describes a method for characterizing olive oil using Agilent GC/Q-TOF MS and Mass Profiler Professional software. The method can detect a large number of compounds in olive oil and perform quantitative analysis.

Solid-phase extraction (SPE) coupled with high-performance liquid chromatography (HPLC) was optimized for extraction and quantification of two fungicides (carbendazim and thiabendazole) in apple juice. Results indicate that SPE using Agilent Bond Elut SCX (60 mg, 3 mL) and HPLC using an Agilent ZORBAX Eclipse Plus C18 column (4.6 mm × 100 mm, 3.5 µm) is an excellent combination for extraction and analysis of these compounds. Recoveries ranged from 92.1 to 99.4 percent with RSDs below 5 percent and limits of detection of 4 µg/kg.

This application note describes the optimization of a rapid LC/MS/MS method for vitamin B related compounds (4-aminobenzoic acid, nicotinamide, riboflavin, and nicotinic acid) on an Agilent ZORBAX Rapid Resolution High Definition HILIC Plus column using an Agilent 1290 Infinity LC System with an Agilent 6410A Triple Quadrupole Mass Spectrometer. The method uses isocratic elution with an acetonitrile rich mobile phase and an ammonium acetate buffer. All analytes have good peak shape with this hydrophilic interaction chromatography (HILIC) application. The method is then used to demonstrate the importance of choosing an appropriate injection solvent when performing LC analyses in HILIC mode. Various injection solvents are investigated, including water, acetonitrile, methanol, and combinations. Of the solvents explored, it was determined that pure acetonitrile yields the best retention and peak shape for all compounds, with water being the worst solvent for analytical performance and peak shape.

This application note uses Agilent ZORBAX RRHD columns with UHPLC/MS to compare Morphine’s HILIC and RPLC. Both methods use acetonitrile/ammonium acetate buffer mobile phase for isocratic elution. The results show that HILIC mode allows for more sensitive MS detection due to more efficient spraying and desolvation in the ESI-MS source, as a result of the volatile acetonitrile rich mobile phase used.
