This Application Note describes the separation of a trypsin-digested protein sample, using the Agilent 1290 Infinity LC System with Agilent ZORBAX Eclipse Plus C18, 150 mm × 2.1 mm, 1.8 µm particles column. Peak capacity variation with change in flow rate and time for various gradients were evaluated. Increase in flow rate from 0.2mL/min to 1.2mL/min effectively improved the peak capacity value to double and increase in gradient time from 5minutes to 50 minutes enhanced a 3.4 fold improve- ment in peak capacity value. The unique design of the Agilent 1290 Infinity LC System tolerates 1200 bar column back pressure, allowing higher flow rates, and reducing the run time.

This document introduces a fast method for analyzing environmental phenols by using Agilent Poroshell 120 EC-C18 4.6 × 100 mm, 2.7 µm column, which transfers the method originally developed on a 4.6 × 100 mm, 5-µm column. The gradient and flow rate are scaled while maintaining a constant retention index in order to determine the optimal flow rate for each evaluated column. Simple guidelines for transferring a method are provided. The new separation reduces the analysis time from 20 minutes to less than 3 minutes with a higher peak capacity, and should not require any changes to sample preparation since both columns use 2 µm frits. The method is further scaled to a 50 mm column that produces slightly less resolution than found with the 100 mm column, but with an analysis time of only 2 minutes with an equivalent peak capacity to the original method. While pressure is increased from the original method, it is still below 400 bar and can be easily transferred to any HPLC system.

A rapid, economic, sensitive and reliable method for the determination of epichlorohydrin (ECH) in drinking water is presented. After extraction by methylene chloride, trace-level ECH was quickly concentrated with K-D concentrator. An Agilent 7890A GC System with an Agilent J&W DB-5ms Ultra Inert column were used for ECH analysis. The calibration curves were linear over the range of 0.08–1.60 ng with correlation coefficients greater than 0.9997. Recoveries of ECH at spiked levels of 1.0, and 5.0 µg/L were 104.7% and 107.0%, respectively. The RSD value for ECH reproducibility was lower than 2.21%. The detection limit was less than 0.07 µg/L, which is below the maximum residue limit (MRL) of 0.1 µg/L in European Union regulations for ECH in drinking water.
