Agilent's high efficiency GC columns can be used for PAH analysis. This article introduces the performance of two Agilent J&W columns, DB-EUPAH and DB-5ms. Both columns can separate PAHs well.

This Application Note illustrates the possibilities of the Agilent 1290 Infinity LC system and the Agilent 6460A Triple Quadrupole Mass Spectrometer for quantitative analysis of poten- tial genotoxic impurities (PGIs) in active pharmaceutical ingredients (APIs). A generic approach was used to determine arylamine and aminopyridine PGIs in selected APIs. The influence of column length and mobile phase composition on the recovery of these impuri- ties in real samples were investigated. High resolution UHPLC using a 150 mm RRHD col- umn packed with 1.8 µm particles resulted in the best overall performance. In cases of good separation between API and the target solutes, analysis speed and sample throughput could be increased using a 50 mm RRHD column. Detection limits were below 20 ppb (ng/g, relative to the API) using MS/MS and below 100 ppb (ng/g, relative to the API) with DAD for 2,6-dichloroaniline, which could not be detected with ESI-MS at trace levels.

This Application Note describes a method for the long-term detection of anabolic steroid metabolites in urine using both the Agilent 6890N network GC system with an Agilent 5975B Series GC/MSD and the Agilent 7000 Series Triple Quadrupole GC/MS. Monitoring the sulfate conjugates of these metabolites, instead of the routine glucuronide metabolites, allowed detection of methenolone for a post-administration period that was almost twice as long. This was achieved using GC/MS in SIM mode. Performing the analysis on the Triple Quadrupole GC/MS in SRM mode extends the detection period even further, to almost three times as long as the conventional glucuronide approach.

This study used an Agilent HPLC-Chip coupled with an Agilent Accurate-Mass Q-TOF LC/MS to analyze the glycopeptide and glycan moiety of a monoclonal antibody (mAb). The tryptic peptide map of the mAb was generated and the glycopeptide was assigned using accurate mass measurement. The enzymatically released glycan moiety from the mAb was extracted and analyzed on an HPLC-Chip coupled with a Q-TOF. Diagnostic oxonium ions from a glycan moiety were further used to identify the glycopeptide and the glycan. The superior mass accuracy of the LC/MS platform, combined with the powerful data processing capabilities of Agilent MassHunter and BioConfirm software, enabled identification of the glycopeptides and glycan moiety attached to the protein complex.

This Application Note describes the separation of nine food additives, acesulfame K, aspartame, caffeine, benzoic acid, sorbic acid, saccharine, trans-cinnamic acid and vanillin using the Agilent 1290 Infinity LC system. The ability of the Agilent 1290 Infinity LC pump to deliver mobile phases at high pressures of up to 1200 bars enables separation of the nine selected additives within a run time of 1 min. The Agilent 1290 Infinity Diode Array Detector (DAD) equipped with Agilent’s proprietary Max-Light flow cell provides sufficient sensitivity for reliable analysis of even 1 µL of the sample. The developed method was applied for the screening of additives present in soft drinks and colas.
