This Application Note describes the development and validation of a method for simultaneous determination of paracetamol, diclofenac and ibuprofen using the Agilent 1120 Compact LC. The method has excellent precision, accuracy and low limits of detection.
This document presents an application note describing the development and validation of a liquid chromatography method for analyzing pharmaceutical compounds. The study utilizes the Agilent 1120 Compact LC system to evaluate precision, linearity, and limits of detection for paracetamol, diclofenac, and ibuprofen.
The validated method demonstrates excellent performance with retention time precision under 0.07% RSD and area precision under 1.0% RSD. The analysis achieves high linearity with correlation coefficients greater than 0.9999 and limits of detection ranging from 13 to 298 pg for the analyzed compounds.
| Retention time precision | < 0.07 % RSD |
|---|---|
| Area precision | < 1.0 % RSD |
| Linearity coefficient | > 0.9999 |
| Limit of detection (LOD) | 18 — 298 pg |
| Flow rate | 1.5 mL/min |
| Injection volume | 3 pL |
| Column temperature | 40 °C |
| Detection wavelength | 230 nm |
The total analysis time, including run and equilibration, was limited to 10 minutes.
The method showed excellent linearity with correlation coefficients between 0.999910 and 0.999975.
