This Application Note introduces the concept of peak capacity and investigates the effects of gradient parameters such as flow rate, gradient time, column length and gradient slope on peak capacity. The results show that flow rate and gradient time have a significant effect on peak capacity. The effects of column length and gradient slope on peak capacity are relatively small.

This document describes the use of the Agilent 1260 Bio-inert Quaternary LC system for sensitive and rapid LC methods to characterize the therapeutic protein, P128. Optimized peptide maps were constructed using sub-2-µm column technology to decrease analysis speed for this protein. Size exclusion chromatography (SEC) and ion exchange chromatography (IEX) were performed using the new Agilent Bio-HPLC columns to detect potential impurities or isoforms of the protein preparation. The bio-inertness and corrosion resistance of the instrument coupled with the simple and reproducible methods make this solution suitable for routine use in QA/QC labs.

This Application Note describes the development of an immunoprecipitation method in combination with microfluidic on-chip analysis on the Agilent 2100 Bioanalyzer for sensitive, selective and quantitative detection of endogenous β-galactosidase in crude E. coli lysates. The effects of antibody selection and the importance of preclearing steps for sensitive and specific detection of the target protein are discussed. Quantification of β-galactosidase was achieved via an external calibration curve based on various spiking levels of pure β-galactosidase into E. coli lysate. Results proved the described approach to be quantitative over orders of magnitudes and highly reproducible.

This application note demonstrates the capabilities of the Agilent J&W CP-COX column with the Agilent 490 Micro GC, including separation of permanent gases and backflush possibilities to ensure extended column lifetimes.

Two samples consisting of various nonsteroidal anti-inflammatory drugs (NSAID) are separated using methods developed on an Agilent 1200 Series Method Development Solution controlled by ACD/Labs AutoChrom for ChemStation software. The chromatograph while under control of AutoChrom can screen up to seven columns, thirteen buffers and two organic solvents with columns held in four temperature controlled zones. The software helps plan the next best experiment to perform, allowing the analyst to focus on quickly developing methods using conditions with the best likelihood of success. In this work four columns (Agilent ZORBAX StableBond SB-C18, Agilent ZORBAX Eclipse Plus C18, Agilent ZORBAX Bonus RP and Agilent ZORBAX Eclipse Plus Phenyl-Hexyl) are screened using five mobile phase modifiers. Twenty solvent column experiments are initially screened for each sample set.
