This document describes two methods for analyzing glyphosate, one is pre-column derivatization and the other is post-column oxidation.
This document presents an application note comparing two methods for analyzing glyphosate and its metabolite, aminomethyl phosphonic acid (AMPA), using High Performance Liquid Chromatography (HPLC). The analysis utilizes Agilent HP 1050 Series HPLC modules, including an autosampler, quaternary pump, and fluorescence detector. The study evaluates precolumn derivatization with fluorenylmethyl chloroformate (FMOC) against post-column oxidation and derivatization with o-phthalaldehyde (OPA).
| Excitation wavelength (Precolumn) | 266 nm |
|---|---|
| Emission wavelength (Precolumn) | 305 nm |
| Cut-off filter (Precolumn) | 280 nm |
| Excitation wavelength (Postcolumn) | 230 nm or 338 nm |
| Emission wavelength (Postcolumn) | 450 nm |
| Column type | SAX-300 100 x 4.6 mm id |
| Detection limit (Automated) | approximately 1 ppb |
The detection limit is approximately 1 ppb for each compound.
The HP 1050 Series autosampler with variable-volume auto-injector is used.
The glyphosate is derivatized with o-phthalaldehyde (OPA) reagent.
