About this manual

This document presents a study on the quality assurance of RNA isolated from tissue samples obtained via laser microdissection using the PALM MicroBeam System. The primary focus is on the feasibility of analyzing low-concentration RNA samples (down to 200 pg/µl) using the Agilent RNA 6000 Pico LabChip kit, which was specifically designed for such small sample volumes.

The study investigates the influence of different RNA extraction kits and histological staining procedures on RNA quality and yield. Results indicate significant variability between kits, with extraction/precipitation-based methods generally showing higher recovery than column-based methods. Additionally, Methylgreen staining yielded the best RNA quality and highest yield compared to Hematoxylin/Eosin, Nuclear Fast Red, and Methylene Blue stains.
