This article introduces a target enrichment technology based on Agilent DNA microarrays, which can be used to resequence multiple discrete genomic regions of interest.
This document details a study validating the use of Agilent DNA microarrays for target enrichment in next-generation sequencing. The authors collaborated with Dr. Greg Hannon's laboratory to capture exonic regions relevant to a breast cancer sequencing study, targeting approximately 0.025% of the human genome using an Agilent 244K array.
The study demonstrated that the method is effective, achieving a 2,700-fold enrichment of sequencing reads within targeted regions. The results confirmed that the system covers over 99.8% of targeted regions and provides a normalized average per-base read depth of 27 per million 32-base reads.
The study targeted approximately 1,287 discrete genomic regions.
The normalized average per-base read depth was 27 per million 32-base reads.
