About this manual

TaqDNA Polymerase SL2601 is a recombinant enzyme derived from Thermus aquaticus YTI, offering 5'→3' polymerase and 5'→3' exonuclease activities. It is supplied as 1000 U of enzyme (5 U/µL) in a 200 µL volume and is suitable for routine PCR, cloning of short fragments, and 3'‑end labeling.

The enzyme should be stored at –20 °C in the provided buffer (20 mM Tris‑HCl pH 8.0, 100 mM KCl, 0.1 mM EDTA, 1 mM DTT, 0.5 % NP‑40, 0.5 % Tween‑20, 50 % glycerol) for up to two years. Recommended reaction conditions include 50 mM KCl, 10 mM Tris‑HCl pH 8.6, 0.1 % Triton X‑100, and 1.5 mM MgCl₂, with 0.5–1 µL enzyme per 50 µL reaction. This manual is written in Chinese.

Frequently asked questions

What amount of TaqDNA Polymerase should be used in a 50 µL PCR reaction?

Use 0.5–1 µL of the enzyme (5 U/µL) per 50 µL reaction for optimal performance.

How should the enzyme be stored and for how long is it stable?

Store the enzyme at –20 °C; it remains stable for up to two years.

What defines one unit of enzyme activity?

One unit is the amount of enzyme that catalyzes incorporation of 10 nmol dNTP into an insoluble precipitate within 30 minutes at 74 °C.
