Confirmation of THC in Oral Fluids Using High-Resolution 2-D GC/MS
Oral fluids are increasingly used as an alternative matrix to urine for drug testing. This document details a method to detect drugs of abuse, specifically THC, at concentrations down to 0.5 ng/mL in diluted oral fluid using high-resolution two-dimensional gas chromatography/mass spectrometry (2-D GC/MS). The technique overcomes interference problems from complex sample matrices that often complicate standard analysis.
The described system utilizes a standard quadrupole GC/MSD configuration enhanced with a Deans switch and an air-cooled focusing trap. Samples are separated on a nonpolar DB-1ms column, then heart-cut to a polar DB-17ms column for further separation. This approach provides the required selectivity and sensitivity comparable to MS/MS techniques while remaining simpler and less expensive.
The confirmation technique must detect THC down to 0.5 ng/mL of diluted oral fluid.
