This document introduces the method of using ZORBAX 300SB analytical column for protein separation. By optimizing the method, a quick and easy scale-up from analytical to semi-preparative HPLC can be achieved.
This document describes a method for developing preparative High-Performance Liquid Chromatography (HPLC) methods by scaling up conditions from an analytical column. The authors demonstrate that using identical packing materials allows for straightforward optimization of gradient separations for protein mixtures, such as Ribonuclease, Lysozyme, Cytochrome C, and BSA.
The note details the specific conditions used on a 4.6 x 250 mm analytical column and provides calculations to determine flow rates for a 9.4-mm inner diameter semipreparative column. It highlights that this approach minimizes solvent usage during method development while maintaining resolution when moving to larger scale columns.
Gradient conditions can accommodate an injection volume of up to 100 µL, which corresponds to a loading of 1 mg per protein.
The calculated flow rate for the semipreparative separation is 4.18 mL/minute to maintain constant linear velocity.
A total of 80 mg of total protein was successfully loaded and separated on this semipreparative column.
