This Technical Overview shows the development of a chromatographic method for the analysis of paracetamol and its impurities using 1290 Infi nity LC System with ISET. Having developed the method it was transferred to an Agilent 1100 Series Quaternary LC System.

This Technical Overview discusses the compatibility of Agilent Jet Stream thermal gradient focusing technology with CE/MS. The combination of capillary electrophoresis and mass spectrometry using the Agilent triple tube coaxial sheath-flow sprayer is widely used due to its simplicity, versatility, and robustness. The study shows that with the Jet Stream source and the latest version of the CE/MS sprayer, this approach can now also be used in combination with the proprietary Agilent Jet Stream thermal gradient focusing technology. The sprayer performed well on a mass spectrometer equipped with a conventional ESI source and showed slightly improved performance compared to the standard sprayer. When the sprayer was used with an Agilent Jet Stream source, equivalent or better results were obtained compared to a conventional ESI source.

This document details the performance characteristics of the Agilent 1200 Series LC system, including performance evaluation steps, module-specific and complete system performance evaluation methods, and sample selection related to system evaluation. Different separation methods were used to evaluate the module-specific and complete system performance. The results showed that the performance of the pump, autosampler, and detector are related to the system configuration, while baseline noise and precision of retention times and peak areas are affected by the complete system.

This document introduces the Agilent 1200 Infi nity Series High Dynamic Range Diode Array Detection Solution, which provides a 30× wider linear dynamic UV range. By combining the signals from two diode array detectors with different path length Max-Light fl ow cells, the high dynamic range DAD solution detects and quantifies main and trace compounds in a single run without exceeding the linear UV range. Using a conventional DAD, typically two injections with different injection volumes are needed to determine the low dose and high dose as well as the trace compounds. At the high injection volumes that are required to determine the low dose and trace compounds, the high dose compounds exceed the linear range of a conventional detector. This Technical Overview demonstrates that one single injection is adequate to reliably quantify low dose, high dose, and trace compounds of a fixed dose combination drug using the high dynamic range detection solution from Agilent. The results are compared to those from a conventional DAD. In addition, the precision of areas and limit of detection for both detection solutions are evaluated and compared.

The Agilent Automated Card Extraction (AACE) LC/MS System is a fully integrated instrument for the automated analysis of Dried Blood Spots (DBS) and other dried media. The system consists of the AACE instrument which enables automated flow-through analysis of DBS cards, together with software to control online analyte extraction, trapping, and LC/MS analysis. The AACE instrument is a CTC-based autosampler platform which integrates with the Agilent 1200 Infinity Series HPLC to provide analytical LC separation. Quantitative analysis is performed using any of the Agilent 6400 Series Triple Quadrupole LC/MS Systems. Data processing and reporting is performed using MassHunter software.
