This application note describes the applications and experimental considerations for FastMAS and UltraFastMAS techniques. FastMAS and UltraFastMAS techniques enable experiments with rotation speeds of 40 to 65 kHz, which reduces linewidths and suppresses spin diffusion caused by proton-proton dipolar interactions. These techniques enable experiments such as direct proton observe or solid-state HETCOR to collect spectra comparable to those that formerly required both spinning and experimentally difficult multiple-pulse homonuclear decoupling sequences. Another advantage of FastMAS and UltraFastMAS techniques is that they suppress spin diffusion between protons, allowing experiments to be conveniently performed on individual protons in single or multiple dimensional experiments.

This document describes the enhanced analysis of semi-volatile compounds using sub-ambient ELSD and HPLC. Evaporative light scattering detection (ELSD) is a powerful tool that can detect any sample that is less volatile than the mobile phase in HPLC applications, regardless of the optical properties of the compounds. ELSD offers distinct advantages over conventional UV or DAD detection, especially for gradient separations. In order to maximize the detection of non-UV absorbing semi-volatile analytes, ELSD detectors need to operate at lower temperatures. Most ELSDs today can evaporate highly aqueous eluents at ambient temperature, but for low molecular weight, low boiling point compounds, ambient temperature is still too high and sensitivity is poor. Sub-ambient ELSD is required to improve the detection of these compounds. The Agilent 385-ELSD is specifically designed to operate under these demanding conditions. It features a Peltier cooled evaporation tube that provides sub-ambient ELS detection down to 10 °C. Dry nitrogen gas is added during the evaporation step to facilitate solvent removal at sub-ambient temperatures.

This file describes the chiral separation and enantiomeric excess (EE) determination of three model chiral compounds using Supercritical Fluid Chromatography using the Agilent 1260 Infinity Analytical SFC system. The low detector noise of the system due to the SFC pump technology resulted in excellent sensitivity for trace analyte detec- tion of chiral impurities. When applying method optimization sufficient resolution was achieved for all compounds even under mass overload conditions. This allowed for the quantitation of impurities at 0.05 to 0.1% levels.

This document describes the method of using the Agilent 1260 Infinity Analytical SFC system for fast and ultra-fast SFC analysis. By using high flow rates, sub-2-µm particle technology, and short columns, separation times of less than 30 seconds with good resolution can be achieved.
