This application note describes a method for identifying oxidation sites on a monoclonal antibody (mAb) using an Agilent 1260 Infinity HPLC-Chip/MS system coupled to an Agilent Accurate-Mass 6520 Quadrupole Time-of-Flight (Q-TOF) LC/MS. A monoclonal antibody was oxidized by treatment with hydrogen peroxide to stimulate potential oxidative modifications that may occur during the manufacturing process. The accurate mass measurement of the 6520 Q-TOF LC/MS combined with Agilent MassHunter BioConfirm software facilitated easy identification of the oxidation sites. Peptides containing amino acid residues such as methionine and tryptophan, which are sensitive to oxidation, were easily identified using this approach.

A method for simultaneous determination of 11 β2-agonist residues of clebuterol, salbutamol, ractopamine, terbutaline, salmeterol, propranolol, tulobuterol, cimaterol, mabuterol, mapenterol, and zilpaterol in pork has been developed and validated. The analytes are extracted by liquid-liquid extraction (LLE) and solid-phase extraction (SPE) and quantified by liquid chromatography coupled to electrospray ionization tandem mass spectrometry (LC-ESI-MS/MS) operating in positive ion multiple reac- tion monitoring (MRM) mode. The method provides a pg/g level of limit of detection (LOD) for all β2-agonists in pork. The dynamic calibration ranges for these compounds are obtained from 0.25 to 5 ng/g. The overall recoveries range from 82% to 105% with RSD values between 1.6% and 8.4%.

The GC analysis of four common polyunsaturated fatty acid methyl esters (PUFA FAMEs) in algal biodiesel is described using method EN 15779. An Agilent 7890A GC system was configured and calibrated according to the procedure outlined in the method. Two samples of B100 biodiesel made from algae oil were each prepared in duplicate and analyzed according to the conditions set forth in the method. In each sample, the four PUFA FAMEs were chromatographically separated and quantified. The analysis precision was calculated and shown to exceed the specifications of the EN 15779 methods.
