About this manual

ArcticExpress™ Competent Cells are designed to improve the expression of recombinant proteins by utilizing cold-adapted chaperonins from the psychrophilic bacterium Oleispira antarctica. These cells facilitate proper protein folding and increase the production of soluble, potentially active protein at lower growth temperatures (11-13°C), avoiding the formation of inclusion bodies common in standard E. coli hosts.

The technology combines cold-adapted chaperonins Cpn60 and Cpn10 with codon bias correction options. By expressing extra copies of specific tRNA genes, certain ArcticExpress cell lines can also address issues where the recombinant protein's codon usage is inconsistent with the E. coli host, further enhancing expression success.

Frequently asked questions

How do ArcticExpress Competent Cells improve protein solubility?

They utilize cold-adapted chaperonins from the bacterium Oleispira antarctica to facilitate proper protein folding at lower temperatures.

What temperature should be used for optimal growth with these cells?

The document recommends lower cultivation temperatures, specifically between 11-13°C.

Do these cells help with codon bias issues?

Yes, specific ArcticExpress lines contain extra copies of argU, ileY, leuW, and proL tRNA genes to rescue expression restricted by rare codons.
