Confirmation of THC in Oral Fluids Using High-Resolution 2-D GC/MS

This article introduces an integrated approach to improve sequence coverage and protein identification by combining liquid chromatography-matrix-assisted laser desorption ionization mass spectrometry (LC-MALDI MS/MS) and nanoflow liquid chromatography-tandem mass spectrometry (nano-LC/MS/MS). The coupling of RP-HPLC with MALDI via a MALDI spotting device provides an automated and robust solution for analyzing medium complexity samples using MALDI. The experimental results show that this integrated approach can improve protein coverage for complex protein samples and facilitate protein identification.

This document describes Agilent's dual-mode Gene Expression Microarray platform and the method for gene expression analysis using RNA isolated from human whole blood. The platform combines the ease and simplicity of one-color experimental design with the capability of two-color sensitivity for detection of small gene expression changes. Agilent's platform now includes a method for the analysis of differential gene expression from human whole blood, which is compatible with the PAXgene™ Blood RNA System. The PAXgene Blood RNA System consists of a blood collection tube and nucleic acid purification kit for the collection, storage, and transport of blood and stabilization of intracellular RNA for subsequent isolation and purification. Combined with the PAXgene System, Agilent's amplification and labeling technology, in conjunction with the high sensitivity and specificity of 60-mer oligonucleotide microarrays, offer a total solution for gene expression in whole blood without the need for additional steps or kits to remove globin transcripts. Titration experiments using Agilent RNA spike-in control targets demonstrate the sensitivity of detection.

In this work, a collision energy ramp is used in conjunction with an ion trap mass spectrometer to generate reproducible product ion spectra. For the purpose of identifying compounds in real samples, the generation of product ion spectra in those samples is also carried out using ramped collision energy. The process of confirmation includes the generating of compound spectra in the sample data, followed by NIST- based scoring of Fit, Reverse Fit, and Purity matching of library spectra. Searching of the MS spectra is not included because matrix interferences can result in poor scoring. Besides, the precursor ion spectra search is also not included because the search may result in overlapping with other compounds.

This paper presents seven examples of pharmaceutical analyses employing ZORBAX Eclipse Plus C8 columns. Eclipse Plus C8 is an excellent starting point for method development because of its excellent peak shape for bases, acids, and neutrals. Excellent peak shape contributes to better peak resolution. Eclipse Plus C8 exhibits different selec- tivity compared to Eclipse XDB C8 and Eclipse Plus C18. The shorter aliphatic C8 chain is less retentive than a C18 chain and can significantly cut analysis time compared to C18. Additionally, there is a wide variety of column con- figurations available for all analytical, rapid-resolution (RR), and high-throughput analyses. Several of these examples are scaled down from analytical methods to RR methods by substituting the longer column with a RR column. This combination of excellent peak shape and scalability to high-throughput methods make Eclipse Plus C8 a valuable tool to increase productivity wherever HPLC is used.

This article describes an analytical method for the detection of cannabinoids in oral fluid. The method utilizes an Agilent 5975 GC/MS instrument and allows for the simultaneous determination of Δ9-tetrahydrocannabinolic acid A (THCA-A), tetrahydrocannabinol (THC), cannabinol (CBN), and cannabidiol (CBD) in oral fluid samples. Nanospray LC-MS analysis of dextromethorphan (DEM) and its metabolites was performed using the Agilent 1200 Series HPLC-Chip/TOF system. The high sensitivity TOF in combination with automated nanospray HPLC-Chips enabled the detection of all DEM metabolites at concentration levels 100-fold lower than normal. The system response was linear over a wide concentration range for metabolites even when spiked into human plasma and urine. The LODs for real-life samples were in the high attomol to low femtomol range. Sample preparation was simplified using acidic precipitation by perchloric acid and a short centrifugation step. The simplified sample preparation, accurate mass measurements, excellent chromatographic reproducibility, and easy-to-use nanospray chip system make this setup optimal for routine use, allowing for higher throughput and reduced sample preparation costs.
