This Application Note describes the general use of cell fluorescence assays on the Agilent 2100 bioanalyzer. The cell assay extension together with the cell fluorescence LabChip® kit allows both the analysis of pre-stained cells and the analysis of cells by on-chip staining. Cell assays on the 2100 bioanalyzer are based on simple flow cytometric analysis. The technical principles, as well as the typical workflow in sample and chip preparation are described here. A typical dye optimization experiment was performed and the appropriate dye concentration for a specific cell line determined. The cell fluorescence software is easy-to-use and allows different forms of data evaluation and result presentation. Using an example, data evaluation is explained in detail. Specific advantages of cell fluorescence analysis with the Agilent 2100 bioanalyzer are low cell and reagent consumption, ease-of-use, automated data acquisition and evaluation of up to six samples.

This paper introduces a method for the separation of basic drugs, such as ranitidine and paroxetine, based on hydrophilic interaction liquid chromatography (HILIC) and electrospray ionization mass spectrometry (MS/MS) detection. The method showed good linearity over the 0.5–100 ppb levels. A spiked serum was cleaned up by protein precipitation and an aliquot was directly injected into the HILIC/ESI-MS system. Recovery was found to be 98% for ranitidine and 79% for paroxetine at the 100 ppb level.

This application note describes how the Agilent 2100 bioanalyzer can be used for the detection of a point mutation in the human prothrombin gene. The prothrombin protein plays a key role in blood clotting. A single nucleotide polymorphism (SNP), i.e. a point mutation in the prothrombin gene results in a common hereditary predisposition to venous thrombosis. We present a convenient and reliable mutation detection method by a PCR-based assay including a specific restriction digest (PCR-RFLP). Due to its high sensitivity and resolution, the applied microfluidic assay proves to be extremely suitable for detec- tion of the obtained PCR products and restriction fragments. The developed method is suitable for the workflow at Laborigo molecular diagnostic laboratory.
