About this manual

This document provides instructions for the use of the Acid Phosphatase Reagent (Catalogue numbers TR27010, TR27015) manufactured by Thermo Fisher Scientific. The reagents are intended for the quantitative determination of total and prostatic acid phosphatase in human serum on both manual or automated systems using an Alpha-Naphthylphosphate Method.

The kit includes Reagent A (containing alpha-naphthylphosphate), Reagent B (containing Sodium L-Tartrate), and Reagent C (Acetate Buffer). The assay measures the hydrolysis of alpha-naphthylphosphate to liberate alpha-naphthol, which is then coupled with Fast Red TR to form a diazo dye. Prostatic acid phosphatase activity is determined by subtracting the non-prostatic result from the total result.

Frequently asked questions

What is the intended use of this reagent?

This reagent is intended for the in vitro quantitative determination of total and prostatic acid phosphatase in human serum on both manual or automated systems.

How should the sample be stabilized?

Acid phosphatase is unstable at the pH of serum; stabilize by adding 0.020 mL of stabilizer (Reagent C) to every 1 mL of serum.

Is calibration required for this assay?

No, calibration is not required as the rate of reaction is converted to U/L by a calculation factor.

What are the expected values for Total Acid Phosphatase at 37°C?

The expected range is 0 - 6.0 U/L (0.0 - 0.10 pkat/L).
