This article introduces a method for determining the content of homopiperazine in fasudil hydrochloride drugs using ion chromatography. Fasudil hydrochloride is an effective vasodilator that can relieve cerebral vasospasm. Homopiperazine is an intermediate in the synthesis process of fasudil hydrochloride. By using ion chromatography and a conductivity detector, the content of homopiperazine in fasudil hydrochloride drugs can be accurately determined.
Fasudil hydrochloride is an effective vasodilator drug. This document presents an ion chromatography method with conductive detection to measure the impurity homopiperazine, which is difficult to detect using standard reverse-phase liquid chromatography due to low concentration and lack of UV absorption.
The study optimizes sample pretreatment conditions, identifying a 17 mmol/L MSA solution as optimal. It compares different column types (IonPac CS12A, CS16, CS17), concluding that the IonPac CS17 provides the best separation efficiency with minimal tailing. The method achieves a linear range of 0.05-5.0 mg/L for homopiperazine.
| Instrument System | ICS 2100 system (EG eluent generator) |
|---|---|
| Analytical Column | IonPac CS17, 250x4 mm |
| Guard Column | IonPac CG17, 50x4 mm |
| Eluent Composition | 17 mmol/L Methanesulfonic acid + 15% Acetonitrile |
| Flow Rate | 1.00 mL/min |
| Loop Volume | 25 uL |
| Detector Type | Suppressed Conductivity Detector (CMMS 300) |
| Linear Range | 0.05-5.0 mg/L |
| Limit of Detection (LOD) | 0.015 mg/L |
The optimal solution is 17 mmol/L MSA, as concentrations greater than this do not improve peak area.
The IonPac CS17 column is recommended because it has lower capacity and weaker hydrophobic interaction, allowing for quantitative analysis without severe tailing.
The method shows good linearity in the concentration range of 0.05 to 5.0 mg/L.
