This document describes a method for automated sample preparation, retention time locked GC-MS and automated data analysis for metabolomic study
The document presents a scientific evaluation of an automated sample preparation method combined with retention time locked Gas Chromatography-Mass Spectrometry (GC-MS) analysis. The study focuses on the metabolomic profiling of *Arabidopsis thaliana* to assess the reproducibility and stability of derivatisation processes, specifically oximation and silylation.
The experimental setup utilized an Agilent 7693A automatic Liquid Sampler for sample preparation upstream of an Agilent 7890A GC & 5975 MSD. The study analyzed plant tissue samples from two genotypes (Wassilewskija and Columbia) at different ages, extracting metabolites using chloroform/methanol/water. Results demonstrated that automated preparation significantly improved reproducibility compared to manual methods.
The process is used for the metabolomic study of Arabidopsis species to enhance repeatability and keep the time between sample preparation and analysis short and constant.
The sample is mixed at 2000 rpm for 2 times, with each mix lasting 30 seconds.
The vial is heated at 30 degrees C for 3600 seconds in the first step and at 37 degrees C for 1800 seconds in the second step.
