This document describes a new HPLC method for the determination of the tricyclic antidepressant doxepin in 'Aponal 100' tablets. The method uses an extra densely bonded Eclipse XDB-C18 column, which is made with a special Zorbax silica support and designed to reduce or eliminate strong adsorption of basic or acidic compounds. The method is stable at neutral pH and shows excellent and rapid resolution of tricyclic aromatic amines at low buffer concentration. Doxepin is separated from 'Aponal 100' tablets with no interference from excipients.

The document describes the analysis of tricyclic antidepressant drugs. Tricyclic antidepressants are tricyclic derivatives of diphenylamine or diphenylmethane and have mood-enhancing effects. Antidepressants inhibit the mechanism that inactivates these amines, resulting in a higher concentration at the corresponding receptors. They have parasympatholytic side effects that occur a few days after treatment. The desired antidepressant effects occur after two to three weeks.

Quinine is used as an anti-malaria medicine. Some soft drinks, such as tonic water, contain up to 80 mg/l of quinine, which gives them a bitter taste.
