About this manual

The cDNA first‑strand synthesis premix reagents (MRT6104-2, 20 rxn; MRT6104-5, 50 rxn) are high‑sensitivity RT‑Mix formulations designed for the first step of two‑step RT‑PCR. They enable synthesis of first‑strand cDNA from very low amounts of total RNA or poly(A) mRNA without adding separate RNase inhibitors, yielding longer cDNA up to 15 kb.

The datasheet provides detailed component lists, storage conditions (‑20 °C, one‑year shelf life), and step‑by‑step usage instructions, including RNA preparation, primer selection, incubation temperatures, and optional DNase/RNase treatments. Precautions and optional procedures are also described to ensure optimal cDNA yield and downstream PCR compatibility. This manual is written in Chinese.

Frequently asked questions

How should the RNA template be prepared before cDNA synthesis?

Mix the RNA template with the chosen primer and DEPC‑ddH2O, incubate at 65 °C for 5 minutes, then place on ice for 2 minutes before adding the remaining reaction components.

What are the recommended storage conditions for the premix reagents?

Store the reagents at –20 °C; they remain stable for one year under these conditions.

What is the maximum amount of cDNA product that can be added to a PCR reaction?

The cDNA product should not exceed 10 % of the total PCR reaction volume to avoid affecting yield.
