This manual introduces the auto-tuning process of Agilent 5975C series triple quadrupole mass spectrometer, including the concept of tuning target, tuning process, tuning result confirmation method, etc.
The document introduces the automated optimization of ion detection, commonly known as autotuning, for the 5975C Series of mass spectrometers. It details how the system adjusts parameters to meet specific criteria, such as calibrant ion abundances, and explains the role of the electron multiplier (EM) in amplifying signals. A key focus is the shift from traditional voltage-based tuning to a superior approach that adjusts the multiplier voltage to produce a specific GAIN, ensuring better consistency and agreement between different instruments.

Compound responses generally follow the GAIN; for example, ATUNE.U+0V yields the lowest response, while HiSense.U yields the highest.
For trace analysis (analyte amounts < 1ng), the HISense.U is recommended. For higher concentrations (1 ng), lower EM Gains like ATUNE.U are more successful.
High concentrations may saturate the detector, causing flat-topped peaks and collapsing the linear working range, while the signal-to-noise ratio may not improve beyond the HiSense.U setting.
