This application note introduces the superior mass accuracy of the Agilent 6540 Accurate-Mass Q-TOF for high-confidence characterization of unknowns. The capability of the instrument was tested by analyzing a sample described as a compound with a nominal molecular weight of 218 and determining its correct molecular formula.

This application note describes a method for two-channel simulated distillation by gas chromatography (GC) for both hydrocarbons and sulfur. The method utilizes a 7890A GC configured with a high-temperature programmable temperature vaporizer (HT-PTV) inlet and a sulfur chemiluminescent detector (SCD) mounted in series with a flame ionization detector (FID) by use of a special mounting adapter. A simulated distillation (SimDis) software program provides an easy-to-use solution for sulfur and hydrocarbon simulated distillation. The data show that observed boiling point (BP) values agree with the ASTM D2887 consensus BP values within the allowable differences. The system also demonstrates very good repeatability for both hydrocarbon and sulfur SimDis. An example of a light cycle oil (LCO) analyzed according to D2887 is also included.

This application note describes a method for high-resolution analysis of intact triglycerides under non-aqueous reversed phase gradient conditions using the Agilent 1290 Infinity LC System.

The document describes a fast method for the analysis of synthetic antioxidants in vegetable oils. The method uses the Agilent 1290 Infinity LC system equipped with an Agilent ZORBAX Rapid Resolution High Definition (RRHD) column and achieves high throughput in 2 minutes with a backpressure of 1120 bar, which is below the 1200 bar upper limit of the column. Optimization of the mobile phase composition and the temperature are discussed. The figures of merit are illustrated using standard solutions and spiked vegetable oil (sunflower, rapeseed, and olive) extracts. Limits of detection are 1 mg/kg or less in the oil samples. Using a simple methanol extraction, good recovery was obtained for all antioxidants in the oil samples.

High-Throughput LC/MS/MS for cytostatic drugs with an Agilent 1290 Infinity LC System coupled to a tandem mass spectrometer

This document presents a fast method development for salicylic acid process impurities using Agilent ZORBAX Rapid Resolution High Throughput Columns and Agilent 1200 Series Method Development Solution. The method was developed using ACD/AutoChrom software, allowing for screening of multiple columns, buffers, and organic solvents to find the best experimental conditions. The final result was an isocratic separation achieved in under 3 minutes.

This application note describes an approach for automating the biodiesel standard and sample preparation prior to GC analysis using the enhanced capabilities of the Agilent 7693A Automated Liquid Sampler (ALS).

The effects of extra-column volume on 50 and 150 mm columns in 2.1 and 4.6 mm internal diameters were made using a range of capillary tubing volumes installed between the autosampler and column. The results showed that, in using short columns, increases in tubing volume will reduce the column separation efficiency, while increases in tubing volume will have little effect on the separation efficiency of the long column, which is consistent with the results of other scholars.

A precolumn backflush system based on capillary columns using midpoint pressure control is described. Midpoint backflush is made possible with a Capillary Flow Technology (CFT) purged union controlled by an AUX EPC channel on the Agilent 7890A GC system. The key application discussed is prefractionation of crude oil that provides a high resolution separation of the C4 to C12 cut. A general backflush method using Polywax 500 is presented to illustrate the backflush concept.

This application note describes the method development options for separation of vitamin D2 and D3 by UHPLC. Partial separation of vitamin D2 and D3 was achieved by an isocratic LC method using a C18 column and methanol/acetonitrile mobile phase. Several optimization parameters were examined to increase resolution (Rs) including column selectivity and column length. A 2.1 mm × 150 mm Agilent ZORBAX StableBond-C18 column provided the best resolution. Further fine tuning included faster flow rates and mobile phase strength resulting in short analysis times of 3 to 1.5 minutes with Rs of 1.6 to 1.4. The performance of a 3.0 mm internal diameter (id) column was also compared to that of the 2.1 mm id column demonstrating higher sample loading before any loss of resolution in the closely spaced peaks.
