About this manual

The PHA standard (Cat# 2030411) is a lyophilized lectin derived from Phaseolus vulgaris (red kidney bean). It binds to PHA receptors on T‑lymphocytes and induces a non‑specific proliferative response, making it suitable for ELISPOT experiments.

To use, reconstitute the powder with 500 µL sterile PBS, mix, and let stand a few minutes. The resulting solution has a concentration of 25 µg/mL; for a typical well add 10 µL to achieve a working concentration of 2.5 µg/mL (suitable for 10⁵ PBMC). Store the lyophilized product at –20 °C, avoid repeated freeze‑thaw cycles, and use within 12 months. The preparation is filtered through a 0.22 µm membrane and meets endotoxin limits for ELISPOT assays. This manual is written in Chinese.

Frequently asked questions

How should the PHA standard be reconstituted?

Add 500 µL sterile PBS to the lyophilized powder, let stand a few minutes, then mix to obtain a 25 µg/mL solution.

What filtration step is required before using the reconstituted PHA solution?

Filter the solution through a 0.22 µm membrane; it does not adversely affect cell culture.

Is the endotoxin level of the PHA standard suitable for ELISPOT assays?

Yes, the endotoxin content meets the required range for ELISPOT experiments.
