This document describes the AMMS® CD3/CD28 antibody‑conjugated magnetic beads (product code GMP‑TL601) designed for the isolation, activation and expansion of human T lymphocytes, supporting applications such as CAR‑T cell production. It outlines bead preparation, washing, cell‑bead mixing ratios, incubation conditions, and downstream culture steps.
Key procedures include bead washing with PBS containing 1% HSA, mixing beads with cells at a 3:1 ratio (or 1:1 for pure T cells), incubation at 4‑25 °C with gentle shaking, and subsequent culture in IL‑2‑supplemented medium at 37 °C, 5% CO₂ for up to 14 days. Storage, endotoxin limits, and safety precautions are also provided. This manual is written in Chinese.
| Part number | GMP-TL601 |
|---|---|
| Bead concentration | 2×10⁸ beads/ml |
| Applicable species | Human T lymphocytes |
| Storage temperature | 2~8 °C |
| Endotoxin level | <2 EU/ml |
| Shelf life | 24 months |
Mix the magnetic beads with cells at a 3:1 ratio (beads:cells).
For pure T cells, adjust the bead‑to‑cell ratio to 1:1.
Resuspend the beads in a centrifuge tube, add an equal volume of PBS with 1% HSA, vortex for 30 s or invert for 5 min, then place on a magnetic rack to remove the supernatant.
Store the beads at 2 °C to 8 °C.
