About this manual

The BD CBA Non-Human Primate (NHP) Th1/Th2 Cytokine Kit is designed for the quantitative measurement of Interleukin-2 (IL-2), IL-4, IL-5, IL-6, Tumor Necrosis Factor (TNF), and Interferon-gamma (IFN-gamma) protein levels in a single sample. The kit performance has been optimized for analysis of specific cytokines in tissue culture supernatants, EDTA plasma, and serum samples from rhesus, cynomolgus, baboon, or pigtailed macaque non-human primate species.

The assay utilizes six bead populations coated with capture antibodies specific for human cytokines. When mixed with recombinant standards or unknown NHP samples and incubated with PE-conjugated detection antibodies, sandwich complexes are formed. These complexes are measured using flow cytometry to identify particles with fluorescence characteristics of both the bead and the detector, allowing results to be generated in graphical and tabular format.

Frequently asked questions

What is the theoretical limit of detection for this kit?

The theoretical limit of detection is comparable to conventional ELISA, though actual limits may vary slightly due to assay complexity.

Can this kit be used on stream-in-air instruments?

No, it is not recommended for use on stream-in-air instruments like the BD FACStar Plus or BD Influx due to potential signal reduction.

Are the results reported in absolute units?

No, because human-specific antibodies are used to measure non-human primate proteins, measured relative values are reported as pg/mL.
