About this manual

The pCMV-Tag 1 is an epitope tagging vector designed for gene expression in mammalian cells. It allows a target gene inserted into the vector to be tagged with the FLAG epitope (N-terminal, C-terminal, or internal), the c-myc epitope (C-terminal), or both. The resulting constructs can be transfected into mammalian cells and characterized using commercially available tag-specific antibodies.

The kit includes the pCMV-Tag 1 mammalian expression vector, a control vector, and XL1-Blue host strain. Sufficient reagents are provided for 25 reactions and one control reaction. The manual details storage conditions, required additional materials such as T4 DNA ligase and Tag DNA polymerase, and step-by-step procedures for preparing host strains, ligating inserts, transforming bacteria, and verifying the insert.
