The QuikChange Lightning Multi Site-Directed Mutagenesis Kit is a reagent kit for research use. This kit enables multi-site directed mutagenesis.
The QuikChange Lightning Multi Site-Directed Mutagenesis Kit offers a rapid and reliable method for site-directed mutagenesis of plasmid DNA at up to five different sites simultaneously. Using advanced high fidelity enzyme technology, the protocols are accelerated while maintaining accuracy, delivering results up to three times faster than the original multi-site kit.
The process follows a simple three-step protocol: mutant strand synthesis via thermal cycling using a Pfu Fusion-based enzyme blend, digestion of the methylated template DNA with Dpn I restriction enzyme, and transformation into XL10-Gold ultracompetent cells. The total reaction time for synthesis is approximately two hours, followed by an overnight transformation.
| Maximum simultaneous mutation sites | up to five different sites |
|---|---|
| Total reaction time (synthesis) | approximately two hours |
| Enzyme technology | Pfu Fusion-based DNA polymerase |
The protocol allows for multi-site mutagenesis in approximately two hours, plus an overnight transformation.
Catalog #210513 contains enough reagents for 30 total reactions.
You need X-gal and IPTG, along with BD Falcon polypropylene round-bottom tubes.
