The cells are established by NTkmT retroviral vector infection and express polyomavirus middle T antigen. Their endothelial characteristics are confirmed by von Willebrand factor expression and LDL uptake. The cells should be cultured in 90% DMEM + 10% fetal bovine serum at 37°C with 5% CO2.
The cells should be passaged when they reach 85-95% confluence.
The recommended freezing formula is 70% base medium, 20% fetal bovine serum, and 10% DMSO.
Frozen cells should be thawed in a 37°C water bath, transferred to a centrifuge tube, centrifuged at 80g-120g for 2-5 minutes, and then seeded in a 25cm² flask.
