The Biotek ProteoSpin™ kit uses silicon carbide ion‑exchange columns to deplete high‑abundance serum proteins for downstream analysis.
About this manual
The ProteoSpin™ Abundant Serum Protein Depletion Kit provides a fast, antibody‑free method to remove major serum proteins such as albumin, α‑antitrypsin, transferrin and haptoglobin. The kit uses silicon carbide (SiC) spin columns that act as anion exchangers, allowing low‑abundance proteins to bind while salts and high‑abundance proteins flow through.
The protocol includes column activation, sample preparation, protein binding, washing and elution steps performed in a benchtop micro‑centrifuge at 6,700 × g. Each 2 mL spin column can deplete up to 500 µg of serum proteins, and the eluted sample is ready for SDS‑PAGE, LC/MS, 2D‑gel electrophoresis or microarray analysis. Troubleshooting tips and FAQs address common issues such as flow problems, protein degradation, and insufficient depletion.

Frequently asked questions
How much serum can each spin column deplete?
Each spin column can deplete up to 500 µg of serum proteins.
How can I prevent degradation of the eluted protein?
Add 5 µL of Neutralizer to each 200 µL of eluted protein to adjust the pH to neutral.
