This manual describes the use of the GeneMorph II Random Mutagenesis Kit.
The GeneMorph II Random Mutagenesis Kit is a research tool designed for generating amino acid substitutions in proteins through error-prone PCR. It utilizes Mutazyme II DNA polymerase, a blend of two error-prone enzymes formulated to provide useful mutation rates with minimal mutational bias compared to previous versions.
Users can control the mutation frequency (ranging from 0 to over 16 mutations per kb) by adjusting the initial amount of target DNA and the number of amplification cycles. The kit includes sufficient reagents for 30 reactions, including Mutazyme II polymerase, reaction buffer, dNTP mix, and a gel standard.
| Mutation Rate Range | 1-16 mutations per kb |
|---|---|
| Reactions Supported | 30 reactions |
| Storage Temperature | -20°C |
| Mutazyme II Concentration | 2.5 U/ul |
An initial target amount of 500-1000 ng is recommended to achieve low mutation frequencies of 0-4.5 mutations/kb.
High mutation frequencies (>9 mutations/kb) are obtained by using 0.1-100 ng of input target DNA.
Users need a temperature cycler, PCR tubes, and PCR primers.
