About this manual

The EZ‑Sep™ Mouse 1X is a non‑toxic, low‑osmolar iodixanol‑based lymphocyte separation liquid developed by Dakewe for isolating mouse spleen lymphocytes. It provides high purity and yield while being gentle on cells.

The manual details the required sterile equipment, step‑by‑step tissue grinding, centrifugation settings, and post‑separation handling, including optional use for rat spleen or rabbit/rat blood. Special notes address common pitfalls such as mixing of 1640 medium and the separation liquid. This manual is written in Chinese.

Frequently asked questions

Can EZ‑Sep™ Mouse 1X be used to isolate lymphocytes from rat or rabbit blood?

Yes, the same reagent can be used for rat or rabbit peripheral blood after dilution with 1640 medium and following the described centrifugation steps.

Why does the lymphocyte layer sometimes appear very low in the tube?

Because the added 1640 medium mixes partially with the separation liquid, changing the density gradient and causing the cell layer to shift downward.
