Agilent Analysis of color additives in sweets Manual

This study presents a robust reverse phase high performance liquid chromatographic (RP-HPLC) method for the simultaneous determination of 10 synthetic colorants in sweets. Separation and quantification were achieved using an Agilent Poroshell EC-C18 column and an Agilent 1260 Infinity LC System. The robustness of the method was established through partial validation. The suitability of this method for quantifying artificial colorants from a food matrix was demonstrated by analyzing color additives in sweets. Additionally, the HPLC method was successfully transferred to a short Ultra High Pressure Liquid Chromatographic (UHPLC) method using an Agilent 1290 Infinity LC System for faster analysis without compromising resolution. The Agilent 1290 Infinity Diode Array Detector (DAD) was used to select various wavelengths for quantifying different colorants at their absorbance maxima. The limits of detection (LOD), limits of quantification (LOQ), precision, accuracy, and linearity of each colorant were established using both methods. Incorporating the Agilent 7696A Sample Prep WorkBench into the analytical workflow facilitated sample preparation for LOD, LOQ, and linearity studies.
