Agilent 1290 Infinity Thermostatted Column Compartment handbook

The Agilent 1290 Infinity Thermostatted Column Compartment is a device that enables precise column thermostatting over a temperature range from 10 degrees below ambient up to 100 °C, providing infinite flexibility for optimizing the speed and selectivity of a separation. It features ultra-high pressure valves that enable a wide range of solutions such as dual or multi-column selection, sample preparation, or alternating column regeneration. The 1290 Infinity TCC can be used together with 1290 Infinity LC and 1260 Infinity LC systems as well as with existing 1200 Series LC and RRLC or even 1100 Series LC systems. Its features include a temperature precision specification of ± 0.05 degrees for repeatable retention times and peak areas, Peltier cooling and heating for two independent temperature zones from 10 degrees below ambient up to 100 °C, low dispersion optimized for low internal volume contribution, spacious column racks for three 30 cm columns or shorter columns, and unique pull-out and Quick Change Valve design for easy plumbing and valve exchange. It also allows clustering of up to three 1290 Infinity TCC modules for maximum flexibility for multi-method solutions.

This application note describes how the DNA 12000 LabChip® kit can be used with the Agilent Technologies 2100 bioanalyzer to size DNA fragments generated by restriction enzyme digestion. Sizing accuracy was found to be greater than 92 % for all samples analyzed. Compari- son of the chip-based separations to those obtained using agarose gel electrophoresis indicates that the DNA 12000 assay accurately sizes fragments over a wider range than can be usually analyzed on a slab gel.
