The M54HC4538/M74HC4538 is a high performance CMOS bistable multivibrator fabricated in silicon gate C2MOS technology. It has the same high performance as LSTTL combined with true CMOS low power consumption. Each multivibrator features both a negative, A, and a positive, B, edge triggered input, either of which can be used as an inhibit input. Also included is a clear input that when taken low resets the one shot. The monostable multivibrators are retriggerable. That is, they may be triggered repeatedly while their outputs are generating a pulse and the pulse will be extended. Pulse width stability over a wide range of temperature and supply is achieved using linear CMOS techniques. The output pulse equation is simply PW = 0.7 (R)(C) where R is the output resistance and C is the output capacitance.

Agilent RNA quality control using the Agilent 2200 TapeStation system Assessment of the RINe quality metric Manual

This Application Note describes the performance comparison and benchmark of the Agilent 2200 TapeStation system with the Agilent 2100 Bioanalyzer system, the industry standard for RNA quality control. The 2200 TapeStation system performs automated analysis of up to 96 samples with approximate analysis time of 1 minute per sample. This Application Note demonstrates the high correlation between RINe generated by the 2200 TapeStation system with the RIN quality assessment of the 2100 Bioanalyzer system. The high reproducibility of RINe achieved with the 2200 TapeStation system in RNA quality assessment is also shown. The 2200 TapeStation system is, therefore, a reliable system for RNA quality control.

This application note demonstrates the concordance in the results achieved with identical oligonucleotide probes on two versions of Agilent miRNA microarrays [(AMADID 021827, release 12.0) and (AMADID 031181, release 16.0)], based on different revisions of the miRBase database and provided on different microarray formats. Since probes for a given microarray will remain constant for subsequent versions of Agilent SurePrint G3 Human miRNA microarrays, barring any miRBase sequence changes, it is necessary to confirm that the TotalGeneSignal achieved is consistent. The data show Agilent’s strength in probe reproducibility, a crucial factor for identifying miRNA of differential expression.

Agilent Determination of Benzimidazole Fungicides in Apple Juice by Bond Elut Polymer SCX Solid-Phase Extraction with High-Performance Liquid Chromatography Manual

Solid-phase extraction (SPE) coupled with high-performance liquid chromatography (HPLC) was optimized for extraction and quantification of two fungicides (carbendazim and thiabendazole) in apple juice. Results indicate that SPE using Agilent Bond Elut SCX (60 mg, 3 mL) and HPLC using an Agilent ZORBAX Eclipse Plus C18 column (4.6 mm × 100 mm, 3.5 µm) is an excellent combination for extraction and analysis of these compounds. Recoveries ranged from 92.1 to 99.4 percent with RSDs below 5 percent and limits of detection of 4 µg/kg.

Agilent LC/MS/MS of Vitamin B Shows Effects of Injection Solvents with ZORBAX HILIC Plus Manual

This application note describes the optimization of a rapid LC/MS/MS method for vitamin B related compounds (4-aminobenzoic acid, nicotinamide, riboflavin, and nicotinic acid) on an Agilent ZORBAX Rapid Resolution High Definition HILIC Plus column using an Agilent 1290 Infinity LC System with an Agilent 6410A Triple Quadrupole Mass Spectrometer. The method uses isocratic elution with an acetonitrile rich mobile phase and an ammonium acetate buffer. All analytes have good peak shape with this hydrophilic interaction chromatography (HILIC) application. The method is then used to demonstrate the importance of choosing an appropriate injection solvent when performing LC analyses in HILIC mode. Various injection solvents are investigated, including water, acetonitrile, methanol, and combinations. Of the solvents explored, it was determined that pure acetonitrile yields the best retention and peak shape for all compounds, with water being the worst solvent for analytical performance and peak shape.

Agilent Comparing HILIC RPLC of Morphine using Agilent ZORBAX RRHD Columns with UHPLC/MS Manual

This application note uses Agilent ZORBAX RRHD columns with UHPLC/MS to compare Morphine’s HILIC and RPLC. Both methods use acetonitrile/ammonium acetate buffer mobile phase for isocratic elution. The results show that HILIC mode allows for more sensitive MS detection due to more efficient spraying and desolvation in the ESI-MS source, as a result of the volatile acetonitrile rich mobile phase used.

Agilent LC/MS/MS of Fungicides Metabolites in Apple Juice with Agilent Bond Elut Plexa Poroshell 120 Manual(1)

This application note describes an analytical method for determination of carbendazim and 3 other fungicide residues in orange juice with easy sample preparation and LC/MS/MS detection. Developed for the 3 most important systemic fungicides, carbendazim, thiabendazole, and imazalil with its metabolite, this method can be easily expanded to include a broader range of pesticides. A simple and rapid extraction procedure provides clean extracts that without any preconcentration allow for detection of analytes at concentrations below 2 ppb. Quantification was performed using triphenyl phosphate as an internal standard. Excellent accuracy (% recovery) and precision (% CV) data were obtained for three concentration levels: 10, 50, and 250 ppb of each analyte.

Agilent Optimizing Performance of an Agilent ZORBAX RRHD Eclipse Plus C18 Column by Enhancing an Agilent 1290 Infinity LC System for Ultra-Low Dispersion Manual

An Agilent 1290 Infinity LC System is optimized for the lowest possible extra-column volume, to ensure minimal band broadening and optimal results from a small dimension, Agilent ZORBAX RRHD Eclipse Plus C18, 2.1 × 50 mm, 1.8 µm column. Improvements to isocratic and gradient analyses are evident, with the early eluting peaks from the isocratic analysis showing the greatest improvement of up to a 58% increase in efficiency (k’ = 1.6). Gradient analyses are improved by 24 to 32% with respect to conditional peak capacity (k’ = 1.3 to 5.3). Additionally, there is no strong correlation between flow rate and the effect of extra-column volume, with respect to column performance for isocratic and gradient analyses.
